Objectives

The overall aim of WP2 is to provide the physicochemical analysis of the water and a detailed analysis of the sediment.

Measurement of pH, EC, organic matter (OM), organic carbon (C), total nitrogen (TN), NH4+-N, NO3--N, NO2--N, SO42-, total phosphorus (TP) and PO43-P will be performed. In particular, pH and electrical conductivity (EC) will be measured by using the In Situ, Aqua TROLL 400 Multiparameter probe. Analysis of total nitrogen will be performed by the Kjeldahl method and NH4+-N will be measured by employing the indophenol-blue method 63. Total phosphorus will be estimated after persulfate digestion. Nitrates, nitrites, phosphates and sulfates will be determined by Ion Chromatography.

An extended physicochemical analysis will be performed matching the sampling points of the amplicon sequencing approach. Measurement of pH, EC, organic matter (OM), organic carbon (C), ash, total soluble polysaccharides, anthrone C, protein, total nitrogen (TN), NH4+-N, NO3--N, NO2--N, SO42-, total phosphorus (TP) and PO43—P will be performed. Total organic matter, ash and carbon determinations will be performed by dry heating (ashing), as previously described 35. Analysis of total nitrogen will be performed by the Kjeldahl method and NH4+-N will be measured by employing the indophenol-blue method 63. Total phosphorus will be estimated after persulfate digestion. Nitrates, nitrites, phosphates and sulfates will be determined by Ion Chromatography 37. Total soluble polysaccharides and water extractable anthrone-reactive carbon will be estimated as described previously 38. Total phenolics will be estimated by the Folin-Ciocalteu method 39. In addition Scanning Electron Microscopy and/or X-ray Diffraction analysis could be performed in selected sediment samples for elucidating the geochemical environment.
